Is it possible for a ticket created by mail to be automatically assigned to a group according to which e-mail destination it's sent? I have two e-mail addresses registered, one for each department.

Guess its a bit more complicated to understand what you mean.
In both cases the answer is yes. When you have two entities and a mail gets in you can make a rule that decides to which entity the mail should go. But you can also make a rule (if the mail is already in the desired entity) that the ticket should be assigned to a specific user / group. Which way do you want to go?
