I have input the glpi directory under D:\glpi.
Installed PHP
Installed MySQL
Created a new web page in IIS.
Set up the page using port 8540. Could not get it to work on port 80 so I thought I would try something different. Didn't work that way either.
when I go to http://servername:8540, I get an HTTP 400 BAd Request message.
I have gone through and made the changes to the php files according to this site:

Any help on how I can get this working would be greatly appreciated.

If you are set on IIS, that's fine, but I find GLPI sets up a lot easier on Apache, and the Apache system seems a lot better to work with to me. (I tried to use IIS when I first started with GLPI, and it was never anything but trouble.) XAMPP works really well.
As for your issue, it doesn't sound like it has anything to do with the port, if it doesn't work on :80 either.
My Configuration: GLPI .72.3 running on XAMPP from a VMware Virtual Machine (Windows XP). PHP 5.2, Apache 2.2.3...
